A probe-based quantitative real-time PCR assay was developed to detect Tetracapsuloides bryosalmonae, which causes Proliferative Kidney Disease in salmonid fish, in kidney tissue and environmental DNA (eDNA) water samples. We present data that was used to determine assay sensitivity, specificity and to define the limits of detection and quantification.

We developed and validated conventional and quantitative real-time PCR assays for the detection of DNA from the myxozoan parasite Tetracapsuloides bryosalmonae, the causative agent of proliferative kidney disease in fish. Assays were tested on fish tissue and on field-collected water samples to assess diagnostic and environmental DNA capabilities. The specificity, sensitivity, and broad applicability of the present assays surpass previous methods for detecting T. bryosalmonae DNA from fish tissue and water samples.

There is a fundamental knowledge gap on the distribution, prevalence, intensity, and ecology of salmonid myxozoan parasites in the Lake Sammamish watershed, Washington. To address this knowledge gap, we tested water samples for Ceratonova shasta, Parvicapsula minibicornis and Tetracapsuloides bryosalmonae DNA from 84 sites distributed throughout the Lake Sammamish watershed in fall 2019 and 74 sites in spring 2020. Our surveillance identified zones with high waterborne parasite loads and provides a proof of concept for this approach that could be expanded throughout the larger Lake Washington watershed.
Light trap and larval trawl collection of bigheaded carp in the Missouri River Basin, 2016

In the Lamine, Moreau, and Little Chariton rivers, light traps were used to capture larval bigheaded carps moving into tributaries. Traps were set on a weekly basis in each river, at roughly 1-kilometer intervals up to 7 km upstream. Ichthyoplankton tows were done in locations near the confluence of each tributary with the Missouri River, and within the Missouri River itself. Data includes identification of larval fish, size, and developmental information of subsampled invasive carp species. Flow readings were taken at up to five points across the channel with a Marsh-McBirney flow meter, and water quality was measured at the surface with a YSI ProPlus before retrieval of light traps.
