Cardiac abnormalities in patients with mitochondrial DNA mutation 3243A>G
Background Tissues that depend on aerobic energy metabolism suffer most in diseases caused by mutations in mitochondrial DNA (mtDNA). Cardiac abnormalities have been described in many cases, but their frequency and clinical spectrum among patients with mtDNA mutations is unknown. Methods Thirty-nine patients with the 3243A>G mtDNA mutation were examined, methods used included clinical evaluation, electrocardiogram, Holter recording and echocardiography. Autopsy reports on 17 deceased subjects were also reviewed. The degree of 3243A>G mutation heteroplasmy was determined using an Apa I restriction fragment analysis. Better hearing level (BEHL0.5–4 kHz) was used as a measure of the clinical severity of disease. Results Left ventricular hypertrophy (LVH) was diagnosed in 19 patients (56%) by echocardiography and in six controls (15%) giving an odds ratio of 7.5 (95% confidence interval; 1.74–67). The dimensions of the left ventricle suggested a concentric hypertrophy. Left ventricular systolic or diastolic dysfunction was observed in 11 patients. Holter recording revealed frequent ventricular extrasystoles (>10/h) in five patients. Patients with LVH differed significantly from those without LVH in BEHL0.5–4 kHz, whereas the contribution of age or the degree of the mutant heteroplasmy in skeletal muscle to the risk of LVH was less remarkable. Conclusions Structural and functional abnormalities of the heart were common in patients with 3243A>G. The risk of LVH was related to the clinical severity of the phenotype, and to a lesser degree to age, suggesting that patients presenting with any symptoms from the mutation should also be evaluated for cardiac abnormalities.

This dataset was utilized to evaluate associations between accelerated epigenetic and transcriptomic age and NMR-measured metabolic and inflammatory biomarkers. It contains information on biological and chronological age, confounders, and NMR-measured 'omic profiles. This dataset is not publicly accessible because: The data is derived from medical records which cannot be disclosed per federal law (HIPPA) and existing data use agreements. It can be accessed through the following means: The data can be accessed via sending a request to the CATHGEN Executive Committee (https://dmpi.duke.edu/studies/cathgen). An approved IRB application may be required by the committee. Format: The data is tabular data containing columns specifying the epigenetic and transcriptomic age of the individuals along with NMR-measured metabolic biomarkers. Social and demographic confounders are are included within the dataset. This dataset is associated with the following publication: Raab, H., E. Hauser, L. Kwee, S. Shah, W. Kraus, and C. Pacing in congestive heart failure
공공데이터포털
Despite the major advances in medical drug therapy, heart failure remains a syndrome associated with high mortality and morbidity. Biventricular or left ventricular (LV) short atrioventricular (AV) delay pacing is being tested in congestive heart failure patients with left bundle branch block. The aim is to resynchronise the dyscoordinate LV contraction. A number of studies are underway, but it is clear that while some patients respond remarkably, this is highly variable. Accurate identification of patients likely to benefit will be crucial. The mechanism of benefit is unclear. A greater understanding of the physiological consequences of pacing will be necessary to accurately identify these patients.
Global gene expression profiles of cardiac progenitors differentiated from human pluripotent stem cells in 3D culture under simulated microgravity
공공데이터포털
Methods: RNA-seq libraries were prepared using the Illumina TruSeq RNA kit and the TrueSeq method was employed for mRNA enrichment. The libraries were quantified and samples were multiplexed in each lane of the flowcell. Cluster generation was performed and then sequenced on the Illumina HiSeq1000 system. Reads were mapped on the Human Genome Reference and normalized expression table was generated. Results: Among differentially expressed genes 53 of them were up-regulated and 75 were down-regulated. Conclusions: Data demonstrate increased expression of genes associated with growth development and pro-survival in cardiac progenitors cultured under simulated microgravity compared with those cultured under standard gravity. RNA-sequencing analysis was performed to compare global gene expression profiles of cells at differentiation day 8 under simulated microgravity vs. standard gravity.

Short-term PM2.5 exposure and early-readmission risk in Heart Failure Patients
공공데이터포털
In this manuscript EPA researchers used high resolution (1x1 km) modeled air quality data from a model built by Harvard collaborators to estimate the association between short-term exposure to air pollution and the occurrence of 30-day readmissions in a heart failure population. The heart failure population was taken from patients presenting to a University of North Carolina Healthcare System (UNCHCS) affiliated hospital or clinic that reported electronic health records to the Carolina Data Warehouse for Health (CDW-H). A description of the variables used in this analysis are available in the data dictionary (L:/PRIV/EPHD_CRB/Cavin/CARES/Data Dictonaries/HF short term PM25 and readmissions data dictionary.xlsx) associated with this manuscript.
